Background

Evaluating the concentration of samples containing nano-sized particles is a key parameter for researchers in many fields. Izon's SIOS technology platform provides a rapid, accurate, repeatable and cost-effective solution for the analysis of absolute concentration of particles in a fluid sample. The detection methodology of SIOS produces discriminating information at the single particle level and Izon's dynamically adjustable nanopores provide the ability to perform concentration analysis for a wide range of particle sizes.

For researchers involved with the synthesis of all types of synthetic nanoparticles, evaluation of a range of biological samples as well as those working in virology, gene therapy and vaccinology, who require an understanding of viral titre, this capability combined with a concentration measurement range spanning seven orders of magnitude provides a powerful concentration analysis tool.

Utilising Izon's SIOS technology instruments in conjunction with the additional pressure control capability provided by Izon's VPM (variable pressure module) provides a new approach for measurement of sample concentration. This methodology can be applied to all particle types including both charged and uncharged particles in a wide variety of electrolytes, across a wide range of sample concentration values.

Concentration Analysis Methodology

The relationship between an applied pressure and the measured frequency of blockade events is linear for a given particle set (see figure at left). The exact linear response (gradient) varies depending upon the concentration of the particles being measured. For a given applied pressure (x) the blockade frequency response for two different samples are related by the relative gradient values (m) of the response lines. As the particle flux is proportional to concentration this allows the concentrations of the two samples to be directly related through their measured blockade frequency and gradient response over pressure.

Measurement of a standard at a single applied pressure setting provides a reference against which any other sample can be compared to obtain an accurate absolute concentration value.
